The pair - who were married from 2000 to 2005 - were reading the script for 1982 coming-of-age comedy Fast Times at Ridgemont High, and the full thing will be streamed on Facebook and TikTok on 17 September at 9pm ET (18 September at 2am in the UK).
A teaser photo from the charitable reading shows Pitt and Aniston joined by an all-star cast, featuring the likes of Morgan Freeman, Sean Penn, Julia Roberts, Matthew McConaughey and Ray Liotta.
The read is to raise money for CORE, a charitable organisation providing emergency relief amid the coronavirus pandemic, and the Reform Alliance, which aims to 'dramatically reduce the number of people who are unjustly under the control of the criminal justice system'.
In a statement, host Dane Cook said: "We're so excited to finally bring you our Fast Times table read in what I must say is the coolest virtual classroom I've ever been in.
"This amazing group of performers were absolutely committed to supporting two vital organisations - CORE and Reform Alliance - and we're thrilled to share ridiculously fun scenes from our little class project."
Penn - who founded CORE and appeared in the original movie - said: "On behalf of our dedicated team at CORE, I'm so appreciative of the support Dane has given and continues to give to our work.
"I'm always game for a giggle. So to gather remotely for a night of laughs with so many talented folks while providing CORE with much-needed resources to continue our vital work - then consider us fully onboard to play!"
It is not yet known who will play each character, however, we do know that Penn will not reprise his role as Jeff Spicoli.
